研究不同脂肪含量和脂肪替代物对再制片状干酪品质的影响。对5 种不同脂肪含量再制片状干酪的基本理化指标、色泽、质构和感官品质进行测定和评估。结果表明:5 种不同脂肪含量的再制片状干酪pH值、蛋白质和脂肪含量存在显著性差异,酶解浓缩黄油的添加对再制片状干酪亮度值、红度值和黄度值均有明显影响;用酶解浓缩黄油替代全部黄油的减脂50%干酪全质构特性与其他组样品有差异,酶解浓缩黄油部分替代黄油制备的4号减脂干酪全质构特性除凝聚性、回复性外,与1~3号干酪无显著差异;通过感官评定发现,酶解浓缩黄油替代全部黄油的减脂50%干酪(5号)感官评分最低,酶解浓缩黄油部分替代黄油的减脂50%干酪(4号)风味和滋味有所增强,总体得分与全脂干酪产品无显著差异。因此通过降低黄油含量,并添加酶解浓缩黄油可提升减脂再制片状干酪的品质。 相似文献

The effect of low levels of nitrogen, phosphorus and a combination of the two on the distribution of polymeric and monomeric proteins in two high quality spring bread wheat cultivars was investigated for two consecutive seasons. Size exclusion-high performance liquid chromatography (SE-HPLC) was used to determine the quantity and relationships of monomeric and polymeric proteins, and their relationship with flour protein content (FPC) and SDS sedimentation volume (SDSS). The low nitrogen and combined low nitrogen and low phosphorus treatments had a much larger effect on the protein fractions than the low phosphorus treatment alone. The SDS-soluble large monomeric protein fraction and the percentage SDS-insoluble monomeric proteins, were significantly increased under low nitrogen and a combination of low nitrogen and low phosphorus treatments. The percentage SDS-insoluble large and total polymeric proteins was significantly reduced under low nitrogen and a combination of low nitrogen and phosphorus treatments. The SDS-soluble and -insoluble small polymeric proteins were significantly increased under both low nitrogen and a combination of low nitrogen and low phosphorus treatments. The low nitrogen treatment consistently caused the lowest FPC and SDSS values. Two wheat genetic lines (responsive and non-responsive to elevated CO2) grown under ambient and free-air CO2 enrichment (FACE) conditions were compared using fuzzy chromatography mass spectrometry (FCMS) metabolite fingerprinting. A more comprehensive survey of the changes in their chemical composition was made on selected samples using ultra-high-performance liquid chromatography (UHPLC) metabolomic profiling with high resolution accurate mass/tandem mass spectrometry (HRAM/MSn). Principal component analysis (PCA) of the metabolite fingerprints showed four clusters for the two genetic lines (responsive and non-responsive) and the two CO2 levels (ambient and elevated) in score plots. Metabolite profiling of representative samples for each of the four clusters identified 25 and 16 compounds from negative and positive data, respectively, including amino acids, saccharides, phenolic acids, flavonoids, and lipids. Loading plots demonstrated that some saccharides and lipids were responsible for discriminating between not only two genetic lines but also two CO2 levels. Analysis of free amino acids (not bound) showed a clear pattern of reduced concentration for both lines with elevated CO2. After acid hydrolysis, the responsive line 6 (41% increase in yield) showed the same pattern observed for free amino acids, but the non-responsive line 5 (6% increase in yield) showed different trends in concentrations of amino acids with elevated CO2. 相似文献

The sponge and dough mixing process is one of the most common in the world, yet the mechanistic understanding of this process has yet to be sufficiently explored. In this study, aqueous solutions of ethanol, succinic acid, and their combination were prepared at concentrations intended to replicate fermentation times of 3, 4 and 6 h. These solutions were added to a farinograph mixer to make dough using hard wheat, soft wheat, and durum wheat flour. The results indicate that these yeast metabolites (ethanol, succinic acid) impact the mixing resistance, peak mixing value, and dough mixing stability in each of the flour types, likely primarily affected by the ratios of gliadin to glutenin and LMW glutenin in each flour type. Results suggest a stabilizing non-covalent interaction imparted by gliadin at peak mixing time, a stabilizing effect of HMW glutenin during break down, and synergistic effects of ethanol and succinic acid that leads to a faster rate of breakdown in later stages of mixing. It also suggests an increase in mixing resistance when acidulants are added to durum wheat dough. Taken together, this study adds new insights on the sponge and dough mixing process in a way that has not previously been conducted. 相似文献

ROCK promotes high glucose-induced cardiomyocyte apoptosis by inhi-biting PI3K/Akt signaling pathway
AIMTo investigate whether Rho-associated coiled-coil kinase (ROCK) is involved in high glucose-induced apoptosis of primary cardiomyocytes by regulating PI3K/Akt signaling pathway. METHODSPrimary Wistar rat cardiomyocytes were cultured and identified by α-sarcomeric actin (α-SCA) immunohistochemistry. Cardiomyocytes were treated with 5.5, 33 and 40 mmol/L glucose for 48 h. The cell viability was measured by MTT assay, and the mRNA expression of ROCK1 and ROCK2 in the cardiomyocytes was detected by RT-qPCR. Flow cytometry was used to analyze the apoptosis of the cardiomyocytes. The protein levels of ROCK1, ROCK2, cleaved caspase-3, Bcl-2, PI3K, Akt and p-Akt were determined by Western blot. In order to confirm the regulatory effect of ROCKs on PI3K/Akt signaling pathway, the cells were divided into control group (5.5 mmol/L glucose), high glucose group (33 mmol/L glucose) and high glucose+Y27632 (ROCK inhibitor) group. Western blot was used to detect the protein levels of ROCK1, ROCK2, PI3K, Akt and p-Akt. RESULTSAfter 48 h of high glucose exposure, the values of relative cell viability in 33 and 40 mmol/L glucose groups were (79.71±2.43)% and (68.41±7.49)%, respectively, both of which were significantly decreased compared with normal control group (P <0.05). After 48 h of high glucose exposure, the relative mRNA levels of ROCK1 and ROCK2 in 33 and 40 mmol/L glucose groups were significantly increased compared with normal control group (P <0.05). Compared with normal control group, the apoptotic rate in 33 and 40 mmol/L glucose groups was increased significantly (P <0.05). Compared with normal control group, the protein expression of ROCK1, ROCK2 and cleaved caspase-3 in 33 and 40 mmol/L glucose groups was increased (P <0.05), while the protein expression of Bcl-2 was decreased (P <0.05). No significant difference in the protein levels of PI3K and Akt among the 3 groups was observed, while the protein level of p-Akt in 33 and 40 mmol/L glucose groups was decreased compared with normal control group (P <0.05). Compared with high glucose group, the expression of ROCK1 and ROCK2 was decreased in high glucose+Y27632 group. No significant difference in the protein levels of PI3K and Akt among the 3 groups was observed. Compared with normal control group, the protein level of p-Akt in high glucose group was decreased, and the protein level of p-Akt in high glucose+Y27632 group was increased significantly compared with high glucose group. CONCLUSION Under high glucose environment, ROCK may reduce the level of p-Akt by inhibiting the PI3K/Akt signaling pathway, thus promoting the apoptosis of cardiomyocytes. 相似文献

以番茄品种合作903为试验材料,设置0、0.1、0.2、0.4 mmol · L~(-1 )亚精胺(Spd)浸种10 h,研究不同浓度Spd浸种对番茄种子萌发、幼苗生长及高温抗性的影响。结果表明:不同浓度Spd浸种处理均能显著提高番茄种子的发芽指数和活力指数,幼苗地下部干质量和壮苗指数明显提高,其中0.2 mmol · L~(-1 )Spd浸种处理发芽最快,活力指数最高。Spd浸种处理降低了高温胁迫下番茄叶片的相对电解质渗透率、丙二醛(MDA)和H_2O_2含量,其中0.2 mmol · L~(-1 )Spd处理的相对电解质渗透率、MDA和H_2O_2含量最低;抗氧化酶(SOD、APX、DHAR)活性增加,并在0.2 mmol · L~(-1)时达到最大值。综上,0.2 mmol · L~(-1 )Spd浸种能有效促进番茄种子萌发和幼苗生长,提高番茄幼苗高温抗性。 相似文献

为提高土壤含水量预测精度,基于物联网监测数据,提出了粒子群算法(PSO)优化BP神经网络的土壤含水量预测方法。首先应用主成分分析法筛选出影响土壤含水量的关键影响因子,然后构建8-5-1的BP神经网络拓扑结构,应用粒子群算法优化BP神经网络的初始权值和阈值。结果表明:与传统BP神经网络相比,新模型优化了网络结构,避免了陷入局部最优解,具有良好的预测效果;模型的评价指标平均绝对误差、平均绝对百分误差、误差均方根分别为0.259 2、0.010 5和0.135 6,与单一BP神经网络相比,预测精度更高,可满足实际的土壤含水量预测的需要。 相似文献
